Laboratory Best Practices

A focused, practical guide to handling, reconstituting, storing, and documenting research peptides — for qualified laboratory personnel only.

Peptide integrity is determined as much by how a sample is handled as by how it was synthesized. Small deviations in temperature, solvent, or documentation can compromise experimental reproducibility. The following guide summarizes widely accepted practices drawn from major reagent manufacturers and biosafety authorities, adapted to peptide research specifically. All products referenced are for laboratory research use only and are not intended for human or animal consumption.

1. Personal Protective Equipment (PPE)

  • Wear nitrile gloves (powder-free), a buttoned lab coat, and ANSI Z87.1 safety glasses at all times when handling peptide powder or solutions.
  • Change gloves immediately after any suspected contamination and never reuse disposable gloves.
  • For weighing dry lyophilized powder, use a respirator rated N95 or higher and work inside a ventilated enclosure to prevent inhalation of airborne particulates.

2. Reconstitution & Solvent Selection

  • Allow the sealed vial to equilibrate to room temperature before opening to prevent moisture condensation on the peptide.
  • Use bacteriostatic water (0.9% benzyl alcohol) for multi-dose research stocks; use sterile water for single-use applications.
  • Direct the solvent stream gently down the side of the vial — never blast powder directly — and roll (do not shake) to dissolve.
  • For hydrophobic peptides, pre-dissolve in a minimal volume of acetic acid (basic peptides) or dilute ammonium hydroxide / DMSO (acidic peptides) before diluting with aqueous solvent.

3. Aliquoting & Freeze-Thaw Control

  • Divide reconstituted stock into single-use aliquots immediately to avoid repeated freeze-thaw cycles, which accelerate degradation.
  • Label every aliquot with peptide name, lot number, concentration, solvent, and date of reconstitution.
  • Store working aliquots at 2–8°C for short-term use (days to weeks) and at –20°C or –80°C for long-term storage.

4. Storage & Stability

  • Lyophilized powder: store at –20°C in the dark with desiccant; stability is typically months to years under these conditions.
  • Reconstituted solution: store at 2–8°C and use within the documented hold time (commonly 7–30 days, sequence-dependent).
  • Protect light-sensitive sequences (e.g., those containing tryptophan) from UV by using amber vials or foil-wrapped containers.

5. Documentation & Traceability

  • Record the lot/COA number, reconstitution date, solvent used, concentration, and operator initials for every vial prepared.
  • Maintain a storage log (location, temperature, freeze-thaw count) so any degradation trend can be traced.
  • Retain Certificates of Analysis (HPLC, LC-MS, amino acid analysis) and link them to the corresponding experimental records.

6. Aseptic Technique & Contamination Control

  • Reconstitute in a clean, dedicated area — ideally a laminar-flow hood or biosafety cabinet — using sterile pipette tips and solvent-grade water.
  • Swab vial septa with 70% isopropyl alcohol before puncture; never re-enter a vial with a used needle.
  • Avoid touching vial openings, caps, or the inside of pipette tip boxes with gloves or tools.

7. Working Time & Discard Criteria

  • Discard any reconstituted peptide that shows precipitation, discoloration, or turbidity that does not resolve on warming to room temperature.
  • Observe manufacturer-recommended beyond-use dates; when in doubt, verify potency by analytical re-test (HPLC/UV) before use.
  • Never pool leftover aliquots from different reconstitution dates — treat each as a separate lot.

8. Analytical Verification

  • Where possible, confirm concentration by UV absorbance (A280 for aromatic residues, or A205/214 for general peptide bonds) before beginning a study.
  • Use orthogonal methods — HPLC for purity, LC-MS for identity — to verify integrity after long storage or before critical experiments.
  • Track batch-to-batch variability across experiments by recording the lot number with each data set.
